I am looking to buy a customized chopper motorcycle 1971 harley that has no front brakes, just rear. Is it mandatory to have front brakes in the state of Indiana on a motorcyle? Will I not be able to plate it with no front brakes?

A vintage bike that did not have them originally, no. Grandfather clause. A '71 Did. It depends on title. If title says 71 Harley and you have inspections, it won't pass. It would need title as homebuilt and some kind of ASN. Then it would only need one set of brakes on one wheel. It may save your life some day.
On 4 wheels fine on 2 u finish up with only one wheel/tyre, not good, so no plugs / repairs new tyre.
Plugs in motorcycle tires aren't safe.It's ok to use one in an emergency such as being stuck on the side of the road but then the tire should be replaced in my opinion.
I tried searching online Quickest way for you to find out, is call your local dealership or bike repair shop that does inspections and ask them. I'm pretty sure it's not required anywhere, it's just a good idea to have one since they have more power than the rear brakes.
More than likely if it originally came with front brakes installed, then you would need to have front brakes on it in order to plate and ride it. DMV from Indiana could answer your question.
