It can. Larger wheels and tires will have a larger circumference. Your car will travel farther for each revolution to the tire. This can have the same effect as installing a higher gear set in the differential. If your vehicle has enough power to overcome this change, your mileage may improve as your engine RPM will be lower. If you don't have enough power to overcome this change, your mileage may be less as the added resistance may cause your engine to labor.
If the tire circumference is roughly the same as the original tire you might actually get better millage because lower profile tires have less side wall to flex and there fore roll easier so if you are going to a tire like a 255 30 R 20 you should get better mileage than a 245 60 R 16 would. Taller tires will make the engine work harder the engine will turn slower and the speedometer would be off. so your mileage would probably be lower.Be sure to buy a rim that allows good air flow to the brakes the brakes will work better and last longer if they stay cool. Light weight wheels will also improve mileage and handling especially on rough roads. The worst part about extremely low profile tires is the lack of side wall flex makes the suspension work much harder and it will give you a harsh ride. If you live in an area where you get much rain keep in mind the low profile tires have a very short contact patch on the ground and are much more susceptible to hydroplaning.
Generally speaking, the larger diameter wheels are much heavier than those that came on the car from the factory, so the short answer is yes. They also affect the handling and ride of the car as well. My advice is to find someone with a car like yours with a similar sized wheel, and ride along. You might like it, you might not.
