Help!!! How can I access/modify my system files! Permissions buttons Add and Remove greyed out?

i need help I have windows vista. and I know i am the administrator of the computer but i am trying to modify some system32 files (copy and replace) and its tells me access denied!so now im trying to edit the permissions for the folder and i cant becuase the add and remove buttons to add full control are GREYED OUT!!uggggghhh this is so frusterating i need help please! preferably from any of you IT Specialists out there THANKS
