Answer:

It really does not make enough difference. When tires are changed they do it off the bike. when they check it they are normally on the kick stand. Your pressure will never be perfect. Even the temperature will play a role from day to day what shows on your tester.
PSI is not affected by the position, placement or orientation of a wheel and tire. It is a static reading which can only be affected by temperature change, barometric pressure and the properties of the gas used to fill the tire ( in this case compressed air ).
