Well honestly you should replace the stud and nut. However your wheel could come loose due to uneaven toruqe and the brake rotor will become out of round and have vibration issues due to improper torque.
Most devices have a 25% safety factor, it's not a problem in the short term. Long term is different, go to a parts store and buy a replacement as soon as you can. Better safe than sorry.
As long as the other 5 are tight then you are ok for the immediate future but defiantly make it a priority to get the missing one.

Hello, Running a 4 lug rim with 3 lugs is not generally safe. You can run if absolutley needed for very very temporary instances.(emergency ) But generally you don't want to 1. it causes stress on the remaining 3. 2. If 1 of the other 3 get loose because of the extra duty, it can cause your wheel to loosen up and egg out ( think of the shape of a egg ) your stud holes. 3. or just out right break. and i dont think you want your wheel trying to pass you in the other lane as you are driving down the road. If the nut itself is missing they are really cheap ( even from a part store ), or if the stud and nut are missing/broke again they are relatively cheap to fix.. all it will take is a 30 min tire off and beat the new stud into its new home.But i seriously wouldnt recommend driving more than to just get it to a parking stall till it is fixed.
